SYNTHESIS OF L-PROLINE FROM THE RACEMATE BY COUPLING OF ENZYMATIC ENANTIOSPECIFIC OXIDATION AND CHEMICAL NONENANTIOSPECIFIC REDUCTION

摘要
We have developed a new procedure for the synthesis of L-proline from the racemate by means of a coupled system of D-amino acid oxidase and sodium borohydride reactions. In this system, D-proline in the racemate is enantiospecifically oxidized to form DELTA1-pyrroline-2-carboxylic acid, which is immediately reduced to DL-proline. D-Proline is eventually exhausted and L-proline is exclusively synthesized in the reaction mixture with a yield of more than 98%. The proline isolated from the reaction mixture was 100% optically pure.
