CYANOBACTERIAL PICOPLANKTON FROM LAKE CONSTANCE .1. ISOLATION BY FLUORESCENCE CHARACTERISTICS

Unicellular autotrophs ranging from 0.9 to 4-mu-m in diameter were isolated from Lake Constance by plating aliquots of water on solidified mineral medium (BG11). Single colonies were selected for further cultivation using epifluorescence microscopy and fluorescence emission spectroscopy to identify species differing in pigment composition. The cyanobacterial isolates represent three pigment types containing phycobilisomes with either phycocyanin or phycoerythrin as dominant accessory pigment or a newly described, highly fluorescent phycocyanin-like pigment. The differences found in fluorescence emission of isolated clones are discussed with respect to in situ strain identification.
