ACYL-THIOESTERASE ACTIVITY IN DEVELOPING SEEDS OF COCOA

Acyl-thioesterase activity has been examined in plastid preparations isolated from two development stages in cocoa reflecting low (105 days post anthesis, dpa) and high (130 dpa) stearate (18:0) production. When supplied with an equimolar mixture of [C-14]acyl-CoAs, both preparations showed a slight preference for palmitoyl-CoA over stearoyl-CoA and oleoyl-CoA. By contrast, the order of preference for mixed [C-14]acyl-ACPs was oleate > stearate > > palmitate. Again there was no change in selectivity with age of the tissue. The results indicate that during maturation the increased accumulation of stearate in cocoa butter does not reflect the induction of a new thioesterase with increased selectivity towards stearoyl-ACP.
