DETERMINATION OF GLYCEROL IN BACTERIAL-CELL WALL TEICHOIC-ACID BY HIGH-PERFORMANCE LIQUID-CHROMATOGRAPHY

摘要
A high-performance liquid chromatography method for the determination of glycerol in teichoic acid was developed. The phosphodiester linkages between repeating glycerol units were hydrolysed by treatment with 5.8 M HCl at 100-degrees-C for 75 h. The degree of hydrolysis was verified by gas chromatography-mass spectrometry. The hydrolysed samples were applied to an ion-exclusion column and the amount of glycerol determined as the area of the peak at 12.6 min. Using glycerol standards an r2 value of 0.999, a relative standard deviation of 2.2% and a detection limit of 0.1-mu-g were obtained.
