摘要
In this study a new combination of embedding and staining methods was developed which permits the visualization of the lignin distribution in the cell walls at the high resolution level of a TEM. The use of water soluble melamine resin for embedding results in extremely ultrathin sections which can be observed in the TEM without supporting films. The embedding allows a staining, specifically of lignin, with KMnO4 with high reproducibility. A densitometric evaluation of the micrographs reveals a great number of details about the lignin concentration within the various cell wall layers.
