Pre-harvest sprouting is a major problem in Sorghum crops which leads to losses in seed viability and produces important decreases in grain weight. In this paper we aimed to have an insight into the physiological basis of pre-harvest sprouting resistance in this crop by assessing germinability, ABA embryonic content and embryonic sensitivity to ABA during seed development in three varieties presenting contrasting sprouting behaviour: Redland B2 (very susceptible), SC 650 (moderately resistant) and IS 9530 (very resistant). Redland B2 caryopses were able to germinate with high germination indices from early stages of development, while caryopses from IS 9530 did not present germination indices different from 0 until near physiological maturity. SC 650 (moderately resistant) grains presented an intermediate pattern of behaviour, In all three varieties isolated embryos were able to germinate with maximum germination indices from as early as 15 d after pollination (DAP). Differences in grain dormancy level were not paralleled by a consistently different endogenous ABA content throughout maturation. However, when ABA embryonic content was measured in incubated 35 DAP caryopses, ABA level in B2 embryos after 24 h of incubation was found to be less than half that observed in IS 9530 embryos after the same period of incubation. In addition, B2 embryos were found to be 10-fold less sensitive to the inhibitory effect of ABA than embryos from the other two varieties. These results explain to a considerable extent differences in germinability between sprouting-resistant and -susceptible varieties and are consistent with differences in sprouting behaviour.
