LOCALIZATION OF BETA-GLUCOSIDASE IN ROOTS OF CICER-ARIETINUM (L) BY A FLUORESCENCE TECHNIQUE

The localization of .beta.-glucosidases in roots of garbanzo beans [C. arietinum] was investigated by a 1-step fluorescence optical procedure using 4-methylumbelliferyl-.beta.-D-glucoside. The well known azo-coupling test for the localization of glucosidases could not be applied in polyphenol containing plants. The results obtained with the new method were comparable with those described in other investigations and showed that .alpha.-glucosidases were absent from the root tissues investigated. The aryl-.beta.-glucosidases were mainly detected in the outer region of the cortex and quantitatively increased towards the root tip.
