OPTIMUM STAINING OF PCNA IN PARAFFIN SECTIONS IS DEPENDENT ON FIXATION, DRYING, AND INTENSIFICATION

摘要
Variable expression of proliferating cell nuclear antigen (PCNA) with immunoperoxidase staining in formalin fixed paraffin embedded tissue is dependent on fixation length, temperature of section drying, and intensity of substrate staining. This study showed that after 24 hr fixation, the tissue is "overfixed." Optimum staining of PCNA was achieved by drying paraffin sections at low temperatures and by increasing DAB staining with imidazole plus a metallic stain such as osmium tetroxide.
