HIGH-PERFORMANCE LIQUID-CHROMATOGRAPHIC DETERMINATION OF PROSTAGLANDINS-F2ALPHA, E2, AND D2 FROM INVITRO ENZYME INCUBATIONS

The high performance liquid chromatographic separation of prostaglandins F//2// alpha , E//2, and D//2 as p-bromophenacyl esters on a microparticulate, bonded, reversed phase column is described. Detection and simultaneous quantitation of less than 3 mu g of each prostaglandin is possible. The method has been applied to monitor prostaglandins produced from sheep seminal vesicle enzyme preparations and to evaluate antiinflammatory drugs on the basis of their inhibition of the synthetase enzyme. Advantages of this technique over other inhibitor screening techniques are discussed.
