INFLUENCE OF TEMPERATURE ON THE SIEVING EFFECT OF DIFFERENT POLYMER MATRICES IN CAPILLARY SDS GEL-ELECTROPHORESIS OF PROTEINS

Two different sieving polymers, branched (dextran) and linear (poly(ethylene oxide)), were used in high-performance capillary gel electrophoresis to study the effect of temperature on the separation of SDS-protein complexes in the molecular mass range of 14 400-97 400 Da. Migration properties and band broadening of the proteins were studied as a function of column temperature ranging from 20 and 50-degrees-C. Equations have been derived that relate the effect of temperature on the mobility of the solute. Using an isoelectrostatic separation mode, the migration time of the SDS-protein molecules decreased in both instances as temperature increased within the examined time interval. Peak efficiency increased with elevated temperature in the dextran-based column but decreased when PEO was used as a sieving material.
