SEPARATION OF PHOSPHATIDYLCHOLINE AND ITS MOLECULAR-SPECIES BY HIGH-PERFORMANCE LIQUID-CHROMATOGRAPHY

Isocratic separation of phosphatidylcholine (PC) and PC molecular species from a soybean leaf lipidic extract using high-performance liquid chromatography (HPLC) is described. PC separation was achieved within 20 min on a semipreparative silica normal phase column using isooctane/2-propanol/water (40: 53:7 v/v/v). As much as 1.25 mg of PC was obtained after a single injection. Individual molecular species of PC were separated on an analytical C18 reversed-phase column using methanol/0.1 M ammonium acetate (95:5 v/v, pH 7.4). Spectrophotometric detection of PC and molecular species was made at 205 nm. © 1990, American Chemical Society. All rights reserved.
