SELECTIVE CLEAVAGE OF TRITYL PROTECTING GROUPS CATALYZED BY AN ANTIBODY

Monoclonal antibodies have been produced that catalyze the cleavage of trityl protecting groups at neutral pH. These antibodies were induced by immunizing mice with a positively charged tris(4-methoxyphenyl)phosphonium hapten. With a 4,4′,4″-trimethoxytrityl ether substrate, a rate enhancement (kcat/kuncat) of 270 was observed. Several other trityl ether derivatives and even a trityl thioether compound were accepted as substrate. The pH-rate profile of the reaction suggests that the observed rate acceleration is not the result of general-acid catalysis in the antibody binding site, but probably derives from electrostatic stabilization of a positively charged transition state. © 1990, American Chemical Society.
