EXPERIMENTAL AND COMPUTER MODELING OF VIBRATIONAL-SPECTRA OF HETEROGENOUS COMPLEXES OF BIOLOGICAL MACROMOLECULES

Comparative analysis of experimental vibrational (5000-400 cm-1) spectra of nucleoproteins with different ratio between protein and nucleic acid was fulfilled. The spectra of DNA and RNA viruses (bacteriophages T4, Cg, lambda-E. coli spp, tobacco mosaic, carnation mottle, potatoe X and Y viruses, densonucleosis virus, enthomoiridoviruses) and model protein-DNA mixtures with relative content from 1:1 to 20:1 were investigated. Calculated spectra were obtained by computer summarring up of molecule component spectra. The differences between the vibrational spectra of the investigated viruses are determined by the protein/nucleic acid ratio as well as by their biological nature. It has been established that Amide 1,.Amide 2 - bands (1700-1500 cm-1)and bands corresponding to the stretching and assymetric nucleic acid phosphodiether bond vibrations (1000-1300 cm) may be used to determine the content of protein and nucleic acid in nucleoproteins.
