A BIOASSAY FOR CHOLERA-LIKE TOXINS USING HT29 CELLS

摘要
In this article we describe an alternative to the Chinese hamster ovary (CHO) cell elongation assay for detecting cholera-like toxins in biological samples. The assay is based on the observation that cholera toxin (CT) and heat-labile toxin (LT) from Escherichia coli cause the formation of large vacuoles in human colonic adenocarcinoma HT29 cells. The response time of HT29 cells to CT or LT was the same as that of CHO cells but much lower concentrations of CT activity could be detected with the HT29 cell assay. Vacuole formation appeared to be dependent on the presence of the ADP-ribosyltransferase subunit of CT.
