INFLUENCE OF COTYLEDONS UPON SUCROSE METABOLISM IN AXIS OF GERMINATING PEA

Growth and enzyme activities of sucrose metabolism of axes attached to, or excised from, cotyledons of pea (Pisum sativum, L. cv. Douce Provence) were measured during 6 days of germination or incubation on aseptic agar-nutritive medium (mineral nutrients, sucrose). Growth of attached pea axes starts from the first hour on germination medium whereas growth of excised axes starts after a lag period of 4 days. The hydrolyses of sucrose (via acid invertase) and starch (via amylase) in excised axes were higher than that of hydrolysis of sugar via SS pathway which predominates in attached axes. Highest activities of enzymes forming hexose phosphate (GK, FK, UDP-Glc-PPiase) and PPi-PFK or phosphorylase (measured in the direction of starch formation) and highest respirometry were observed in attached axes; conversely, activities of SPS and of enzymes of PPP were 2-fold higher in excised axes. Involvement of osmotic adjustment and negative energetic balance in the relation between the lag phase and the metabolism of sucrose was discussed.
