摘要
A mathematical model has been developed describing non-linear substrate amplification in amperometric enzyme membrane electrodes. The coupled enzyme reactions of myokinase and pyruvate kinase have been used to realize an exponential amplification system. Using photometric analysis, the lower detection limit obtained via lactate dehydrogenase and NADH detection was 1.5 nM ADP and the expected exponential shape of the signal-time dependency has been observed. The combination of myokinase and pyruvate oxidase in an immobilized form with a pyruvate oxidase electrode resulted in an amplification factor of 800.
