ACTIVITY OF IMMOBILIZED YEAST ALDEHYDE DEHYDROGENASE IN A FLOW-INJECTION SYSTEM

A study of the activity of immobilized aldehyde dehydrogenase on controlled-pore glass for the the determination of aldehydes in a flow system is presented. The stability of the immobilized enzyme is shown to be very limited; however, it was improved by reduction of the Schiff base formed during immobilization. The importance of the addition of glycine and mercaptoethanol for the activity of the immobilized preparation was studied in relation to the mechanisms of the catalytic reaction. Inactivation of the immobilized enzyme preparations was related to the presence of oxygen in the carrier stream. Under optimum conditions, a 10-mu-l enzyme reactor was found to be stable for at least 1 month. Linear calibration graphs were obtained over the range 3-mu-M-7 mM in a flow system with a sample throughput of 45 h-1.
