Monoclonal antibodies (MAbs) were produced against gilthead seabream Ig, and their reactivity was demonstrated by immunofluorescence microscopy, flow cytometry, immuno-electronmicroscopy and immunochemistry. Only one of these MAbs (WSI 5) appeared to react with living B cells and was selected for further research. WSI 5 was reactive with the L chain (± 25 kDa) of both forms of Ig found in gilthead seabream serum: a tetrameric (± 760 kDa) and a monomeric (± 190 kDa) molecule. Immunofluorescence reactions on cytocentrifuge slides and cryostat tissue sections showed Ig-bearing lymphocytes and Ig-containing plasma cells in head kidney, spleen and thymus. FACS-analysis of head kidney, spleen and thymus revealed about 22%, 23% and 3% of Ig-surface positive B cells, respectively. Immunogold labelling of living cells showed that Ig was frequently present in small clusters at the external membrane of Ig-bearing and Ig-containing cells. © 1993 by Academic Press, Inc.
