摘要
The technique of "on-column focusing" allows the injection of high sample volumes onto a conventional LC or micro-LC column for improved sensitivity without severe peak broadening effects. For early eluting compounds, the application of this technique provided better results for micro-LC studies than for LC assays. An increase in sensitivity of more than two times was obtained with injection volumes of up to 50-mu-L onto a capillary column. Therefore, the main drawback of reduced detectability in miniaturized systems could be overcome. For the late eluting compounds, this technique could be similarly applied to both types of chromatographic systems.
