Nucleosides were derivatized using the new reagent bis-(trimethylsilyI)-trifluoroacetamide. A series of sRNA minor nucleosides from commercial pseudouridine, 5-methyl deoxyuridine, 3-methyl cytidine, and 1-methyl adenosine were found to have relatively large differences in the retention times as compared with the parent nucleoside using gas-liquid chromatography with 1.5% SE-30 columns. Pseudouridine was discovered to have two distinct components which are believed to represent the α and β stereoisomers. There was no advantage for resolving 6-methyl aminopurine riboside and N-1 methyl adenosine using the trimethylsilyl derivatives on thin layer chromatography. However, thin layer chromatographic methods did enable the separation of nucleosides from their trimethylsilyl derivatives. © 1969, Oxford University Press. All rights reserved.
