THE MECHANISM OF FLOC FORMATION IN PROTEIN PRECIPITATION BY POLYELECTROLYTES

Precipitation by polyelectrolytes is an important method in the processing of protein products. In the present work the mechanism of floc formation, breakup and reaggregation is examined in the system consisting of lysozyme precipitated with polyacrylic acids of varying molecular weight. The protein is first precipitated in a stirred tank under gentle agitation followed by a period of vigorous agitation and then a final period of gentle agitation. Floc size distributions are determined at all the three stages and used to draw inferences concerning the mechanism of floc formation and breakup. These inferences are further examined by measuring the molecular weights of the polyelectrolytes in flocs which have been subjected to agitation and comparing them with their original values. The results support a polymer bridging mechanism with high molecular weight polyelectrolytes and a charge neutralization mechanism with low molecular weight precipitant, A gradual transition between the mechanisms is noted as the molecular weight of the polyelectrolyte is decreased.
