A METHOD FOR SIMULTANEOUS DETERMINATIONS OF COPPER AND IRON LEVELS IN A SINGLE SMALL SAMPLE OF MOUSE PLASMA

The application of the technique using sulfonated bathophenanthroline for the determination of iron levels in plasma showed that considerable interference was caused by the presence of copper in mouse plasma. Modifications of the technique produced a simple method for the routine determination of both copper and iron. The colour reagent-sulfonated bathophenanthroline reacts with both copper and iron to give complexes with optical density peaks at 455 and 535 nm respectively. The complex with copper is unstable in the presence of hydrochloric acid, and any change in optical density at 455 nm after the addition of acid is thus a measure of the amount of copper present. The dissociation of the complex also allows iron content to be estimated without interference caused by the presence of copper. © 1970.
