MONOMER-TRIMER TRANSITION OF HUMAN PLACENTA ESTRADIOL DEHYDROGENASE

Human placenta estradiol dehydrogenase (17β-hydroxysteroid:NAD(P)-oxidoreductase; EC 1.1.1.51) prepared with precautions to prevent subtle denaturation exhibits a concentration-dependent monomer-trimer transition. The kinetic properties of the enzyme show that the monomeric form of the protein can catalyze both net steroid dehydrogenation and substrate-mediated transhydrogenation. In contrast, the trimer catalyzes only substrate-mediated transhydrogenation. The trimerization is demonstrable by analysis of the kinetics, by gel permeation chromatography, and by density gradient ultracentrifugation. © 1969.
