THEORY OF VARIABLE-ANGLE SYNCHRONOUS FLUORESCENCE-SPECTRA

Synchronous fluorescence spectra, acquired by scanning excitation and emission monochromators simultaneously, can be modeled by Gaussian distributions of intensity versus energy (cm-1). The peak location, breadth, and intensity can be predicted for constant offset and variable-angle synchronous spectra in the energy domain; linear transformation and multiplication of the excitation and emission spectra result in the appropriate synchronous spectrum. Similar predictions can be made in the wavelength domain because straight lines on the wavelength excitation-emission matrix (EEM) approximate straight lines on the energy EEM. These predictions should permit analysts to select scan conditions to maximize signal intensity and resolution in multicomponent analysis, based only on excitation and emission spectra of the analytes.
