EFFECT OF SEVERAL RATES OF FREEZING AND THAWING ON VIABILITY OF MYCOBACTERIUM LEPRAE

The influence of the viability of the organisms of various rates of freezing and thawing of suspensions of M. leprae in 10% glycerol and 0.1% bovine serum albumin was investigated, employing Shepard's mouse footpad technique as a measure of viability. Freezing and thawing produced some loss of viability. Rapid freezing was less deleterious than slow freezing, and approximately 75% of the organisms survived rapid cooling to -60°C and overnight storage at this temperature, followed by slow warming. Study of the additional effect of prolonged storage on the viability of M. leprae is planned. © 1970.
