A method is proposed that corrects for variable background signals. This technique has been applied to the analysis of polyaromatic hydrocarbons using high-performance thin-layer chromatography with fluorescence detection. The fluorescent background from thin-layer plates was found to be highly variable, and simple background subtraction yielded imprecise and inaccurate concentration estimates. The method proposed is based on the assumption that variable background signals can be modeled by the abstract spectra obtained from factor analysis of several background spectra, and the appropriate weighting factors can then be calculated by using the adaptive Kalman filter.
