HYPOVIRULENCE AND DOUBLE-STRANDED-RNA IN SCLEROTINIA-SCLEROTIORUM

Five isolates of Sclerotinia sclerotiorum were evaluated for morphological differences in agar culture, virulence on celery petioles, and the presence of double-stranded ribonucleic acids (dsRNA). Four isolates grew rapidly in culture, developed typical colony morphologies, were virulent on celery petioles, and did not contain dsRNA. The fifth isolate (isolate 91) grew slowly in culture, developed an atypical colony morphology, produced significantly smaller lesions on celery than virulent isolates, and contained dsRNA. Isolate 91 was considered to be hypovirulent. Hypovirulence was transmitted from isolate 91 to virulent, recipient isolates by hyphal anastomosis. Recipient isolates grew slowly in culture, developed atypical colony morphologies, were hypovirulent, and contained dsRNA. Initiating cultures from hyphal tips and treating colonies with heat or cycloheximide did not eliminate dsRNA from isolate 91. Hypovirulence in isolate 91 of S. sclerotiorum was associated with the presence of dsRNA.
