A system has been described for the separation of the partially degraded products of an immunoreactive preparation using acrylamide-entrapped enzyme, and a tandem series of ultrafiltration cells, each equipped with a different molecular size retentive filter. This technique of digestion with concomitant ultrafiltration affords a means of readily obtaining immunologically reactive fragments of intermediate size. Verification of the changes in antigenicity was obtained by immunodiffusion and immunoelectrophoresis of the membrane-separated components of the enzyme-degraded mixture. © 1969.
