INTRAMOLECULAR TRANSESTERIFICATION IN THIOPHOSPHATE-ANALOGS OF AN RNA-DIMER

The differences in rates for cleavage of uridylyl(3'-5') uridine and the uridin-3'-yl uridin-5'-yl thiophosphates (R(p) and S(p)-diastereomers) have been measured. The ratios of rates are: k(phosphate)/k(R(p)-thiophosphate) = 1.3; k(phosphate)/k(S(p)-thiophosphate) = 0.78; k(S(p)-thiophosphate)/k(R(p)-thiophosphate) = 1.7 and they were found to be independent of pH in the pH-range 9-12. The results are also discussed in the context of using thiophosphates in kinetic studies of enzymatic systems involving transesterification of phosphodiesters.
