DETERMINATION OF SPECIFIC ACTIVITIES OF LABELED STEROIDS USING GAS-LIQUID CHROMATOGRAPHY AND A FRACTION COLLECTOR

摘要
A method for trapping steroids from gas-liquid chromatography effluents has been investigated. Glass wool was found to be preferable to p-terphenyl and 2,5-diphenyloxazole crystals. It was found necessary to have the temperature of the arm of the fraction collector at not less than 202° to prevent tailing of the eluted steroids. Mixtures of 3H-dehydroepiandrosterone and 14C-progesterone were chromatographed and the 3H 14C ratio of the individually collected steroids was found to be unchanged by chromatography, indicating good resolution. The application of this method to the determination of specific activities of steroids isolated from an in vitro incubation with biological tissue is described. © 1969.
