RECOGNITION OF DNA-SEQUENCES BY STRAND REPLACEMENT WITH POLYAMINO-OLIGONUCLEOTIDES

摘要
A modified 11-mer oligodeoxyribonucleotide 1* of sequence ATTG*AATG*TTT, containing two spermine residues linked to the C2 position of inosine (G*; 2-aminoinosine is guanosine) was synthesized. Its duplex 2.1* structure with the complementary strand 2 melted 25-15 degrees C higher than the natural 2.1 DNA, depending on the ionic composition of the medium. Furthermore, polyacrylamide gel electrophoresis showed 1* to be able to replace 1 from 2.1 in a physiologically relevant medium and some 30 degrees C below the melting temperature of 2.1
