PARALLEL ORDERS OF REACTIVITY IN MURINE CELLS OF MEZEREIN AND PHORBOL ESTERS TOWARD THE MIXED-LYMPHOCYTE RESPONSE AND PROMOTION OF TUMORIGENESIS

摘要
Mezerein and 3 phorbol diesters inhibit the murine mixed-lymphocyte response (MLR). The observed order of activity of these agents is the same as that found for their individual tumor-promoting activity on mouse skin. Mezerein elicits various responses to the same quantitative degree as the most powerful promoter, 12-O-tetradecanoylphorbol-13-acetate (TPA), but it is only weakly active in the mixed-lymphocyte response, mirroring its weak promoting activity in vivo. Tumor promotion is divisible into 2 stages, with TPA able to complete both but mezerein only the 2nd stage. The MLR may be useful for investigations into the mechanism of tumor promotion and is perhaps the 1st in vitro murine system for distinguishing between promoters active in one stage or the other.
