摘要
During the past year, protein engineering has been used to investigate the mechanisms of a variety of enzyme-catalyzed reactions. In particular, considerable progress has been made in understanding the importance of hydrogen-bonding interactions that stabilize otherwise unstable intermediates and influence the reactivity of the substrate in the active site. Protein engineering also has been used to characterize the importance of conformationally flexible active site loops in catalysis.
