BINDING OF CHOLESTEROL TO THE RAT AORTA AND COLLAGEN FIBRES DURING AGEING AND ATHEROSCLEROSIS IN VITRO

摘要
1. The incorporation of C-14-Cholesterol into collagen of tendon and in the aorta of young and old rats was measured during incubation of collagen fibres and aorta in serum with C-14-Cholesterol in vitro. 2. The incorporation of cholesterol into collagen fibres and into the aorta decreases markedly with age. The differences appear during an incubation lasting more than 2 hours. 3. Stepwise extraction of C-14-Cholesterol from young and old collagen fibres showed that cholesterol is bound in young and old fibres in similar strength. 4. The C-14-Cholesterol incorporation into collagen fibres and aorta in serum from old animals decreases markedly in comparison with incubation in serum from young animals. 5. The increased amount of cholesterol in the aorta during ageing cannot therefore be explained neither by the changed properties of the structure, nor by the changed properties of the serum. The effect of both is the decrease in cholesterol uptake in old age. 6. Treatment of young collagen fibres with 0.001 per cent formaldehyde (artificial ageing) increases their C-14-Cholesterol uptake. 7. The C-14-Cholesterol uptake by collagen fibres in vitro did not change in animals fed for 11 weeks by an atherogenic diet.
