DEMONSTRATION OF ADENOSINE TRIPHOSPHATASE IN MITOCHONDRIA OF CULTURED CHICKEN CARDIAC MYOBLASTS AFTER FORMALDEHYDE FIXATION

摘要
Light- and electron microscopic investigations of the localization of mitochondrial ATPase in cultured chicken cardiac myoblasts have been carried out. The following results were obtained: 1. Formaldehyde - freshly prepared from paraformaldehyde - in cacodylate buffer solution containing sucrose is qualified to demonstrate the mitochondria objectively. 2. Contrary to glutaraldehyde this fixative preserves the Mg++-dependent mitochondrial ATPase in a reactive and demonstrable condition. 3. This mitochondrial ATPase was demonstrated by the lead salt method of Wachstein and Meisel (1957) and is localized to the intercristal matrix. © 1969 Springer-Verlag.
