PEROXIDASE SECRETION BY PELLETS OR IMMOBILIZED CELLS OF PHANEROCHAETE-CHRYSOSPORIUM BKM-F-1767 AND INA-12 IN RELATION TO ORGANELLE CONTENT

摘要
The ability of pellets or immobilized cell cultures of Phanerochaete chrysosporium Burds. Wild-type BKM-F-1767 and INA-12 to produce extracellular peroxidases was investigated. Lignin peroxidase (LiP) and Mn(II)-dependent peroxidase (MnP) secretion was increased by immobilizing the fungus on polyurethane foam. Strain INA-12 cultivated as pellets or immobilized cells produced high peroxidase activities more efficiently than strain BKM-F-1767. With immobilized cells, maximal LiP activities of 1296 and 2580 U l-1 were obtained with BKM-F-1767 and INA-12, respectively, after 5 days of cultivation. Other extracellular proteins were stimulated in the same way. Although formed in different ratios, isoenzymes produced by both strains were essentially identical with respect to their physical properties (isoelectric points and molecular weights). Determination of marker enzymes revealed that the enhancement of peroxidase secretion observed with immobilized cells was correlated with a proliferation of endoplasmic reticulum and mitochondria marker enzymes for both strains.
