Monolayers of water-soluble immunoglobulin G (IgG) were prepared and their antigen binding properties were studied. Spectral investigations revealed that IgG molecules were closely packed in the monolayer and that the gross secondary structure of the polypeptide chain has not been changed during the deposition. Langmuir-Blodgett films of IgG bind antigen more effectively than IgG immobilized chemically on the same substrate. These data suggest that the Langmuir-Blodgett technique may be used as a method alternative to traditional immobilization for the development of immunosensors.
