MOLECULAR WEIGHTS OF VIRUSES FROM ISOPYCNIC CENTRIFUGATION WITH SCHLIEREN OPTICS

Good results for molecular weights have been obtained for two viruses, bromegrass mosaic virus and ∅X174, from density gradient equilibrium experiments with the schlieren optics of the analytical ultracentrifuge. The main problem encountered was with density heterogeneity of the virus preparations which gave distorted and broadened bands. This problem was circumvented by a data-fitting procedure to extract the molecular weight from regions of the peak unaffected by additional components. Synthetic data were generated to simulate various amounts of an additional component to determine the effects upon molecular weight determinations. Since no preferential hydration of the virus was found, the two-component equation was found to be sufficient. This may be indicative of exclusion of water from the interior of the virus. © 1969.
