A VERSATILE METHOD FOR IN VITRO PERFUSION OF ISOLATED ORGANS OF RATS AND MICE WITH PARTICULAR REFERENCE TO LIVER

Apparatus to perfuse in vitro isolated rat and mouse liver is described and the advantages of the new method are described. Among the parameters tested to establish the function of the perfused liver were: levels of protein, urea, glutamic-oxalacetic transaminase, glucose, FFA, phospholipids, triglycerides, cholesterol, BSP-test, hemolysis of perfusion liquid, bile volume, concentration of bilirubin in bile, BSP excreted in bile, dry tissue component and levels of protein, glycogen, phospholipids, triglycerides, cholesterol and succinic dehydrogenase in liver tissue after perfusion. Histological and histochemical tests were also performed. The results obtained indicate that the liver can be kept in good conditions for at least 90 min. This method can also be adapted for the perfusion of the isolated lung and possibly of other organs. © 1969 S. Karger AG, Basel.
