STEROL COMPOSITION OF TOBACCO CALLI SELECTED FOR RESISTANCE TO FENPROPIMORPH

摘要
Fenpropimorph, a fungicide used in agriculture, is a strong inhibitor of the cycloeucalenol-obtusifoliol isomerase in plants. Selection experiments, consisting of screening large populations of microcalli derived from UV-mutagenized protoplasts of tobacco for resistance to a lethal concentration (40 mg l-1) of fenpropimorph, resulted in the recovery of seven resistant calli. When grown in selective conditions, the resistant calli accumulated 9-beta,19-cyclopropyl sterols in higher amounts than control calli. After three consecutive passages on a selective medium, some calli may contain up to 99% of cyclopropyl sterols. These results suggest that some of the cyclopropyl sterols may be good surrogates of DELTA-5-sterols in the wild-type and resistant calli, and that phytotoxicity of fenpropimorph is not directly related to the inhibition of sterol biosynthesis in wild-type tobacco tissues.
