FLUOROMETRIC-DETERMINATION OF PYRUVIC-ACID AND ALPHA-KETOGLUTARIC ACID BY HIGH-PERFORMANCE LIQUID-CHROMATOGRAPHY

摘要
A HPLC [high performance liquid chromatography] method was developed for the fluorometric determination of pyruvic acid (PA) and .alpha.-ketoglutaric acid (.alpha.-KG). They were separated by anion-exchange chromatography, reacted with N1-methylnicotinamide chloride (NMN) in the presence of alkali, and heated after the acidification with formic acid to produce fluorophores. By using the optimized conditions for chromatography and the post-column derivatzation, 15 pmol of PA and 75 pmol of .alpha.-KG could be determined. The relative standard deviations of the method were 3.82% and 3.32% for the analyses of 500 pmol each of PA and .alpha.-KG, respectively.
