FRACTIONATION OF RABBIT ANTI-HORSE CYTOCHROME-C .1. SPECIFICITY OF THE ISOLATED FRACTIONS

A method has been devised to fractionate rabbit antibodies to horse cytochrome c by affinity chromatography. Immune serum was passed through a series of affinity columns (immunoadsorbents) each prepared with cytochromes c from closely related species. Antibody sub-populations were isolated by selection of the proper sequence of columns, and displayed unitary stoichiometry of reaction (one Fab' molecule bound per cytochrome c molecule) in fluorescence quenching experiments. The localization of the antigenic determinants towards which these tractions were directed was deduced from the pattern of the cross-reactions of such fractions with cytochromes c of known amino acid sequence. Analysis of these fractions reveals that residues 44 (proline). 60 (lysine) and 92 (glutamic acid) influence the structure of individual antigenic determinants of horse cytochrome c. © 1979.
