Using partition column chromatography, three methylated nucleosides have been isolated from normal human urine. The lyophilized urine was fractionated on a diatomaceous earth column using a formic acid gradient in an ethyl acetate‐2‐ethoxy‐ethanol solvent system. Purification of the major fractions, using repeated paper chromatography, led to the isolation of methylated nucleosides, N2‐dimethylguanosine, 1‐methylinosine, and N2‐methylguanosine. The identity of these nucleosides was established by comparison of their paper chromatographic and electrophoretic mobilities and UV spectra with those of authentic samples. Acid hydrolysis and the orcinol color reaction were in agreement with their identity as nucleosides. Copyright © 1969 Wiley‐Liss, Inc., A Wiley Company
