经前期综合征肝气逆证大鼠模型血清及不同脑区孕酮及氨基酸检测分析

摘要
目的:研究经前期综合征(premenstrual syndrome,PMS)肝气逆证大鼠模型血清、不同脑区中孕酮(progesterone,P)、γ–氨基丁酸(γ–aminobutyric acid,GABA)与谷氨酸(glutamic acid,Glu)的含量变化,探明PMS肝气逆证发病机制与三者含量的关系,并考察药物对其含量影响,探讨药物作用机理。 方法:采用细胞涂片法观察大鼠阴道细胞形态,确定动情周期。挑选动情后期大鼠,以情志刺激多因素法复制PMS肝气逆证大鼠模型,药物白香丹胶囊干预。分别采用放射免疫法(RIA)测定大鼠血清及四个脑区(下丘脑、边缘叶、大脑额叶皮层、海马)P的含量,采用柱前衍生高效液相色谱(HPLC)荧光检测法测定GABA与Glu的含量。 结果:大鼠造模后血清中P、GABA及Glu含量与正常组相比,均显著降低(P﹤0.05,P﹤0.01,P﹤0.01),白香丹给药组大鼠血清中三者含量较模型组相比均显著升高(P﹤0.05,P﹤0.01,P﹤0.01)。下丘脑、边缘叶、额叶皮层及海马四个脑区中,模型组与正常组相比,P含量均显著降低(P﹤0.01),白香丹给药组下丘脑、额叶皮层及海马三个脑区中P含量显著升高(P﹤0.01,P﹤0.05,P﹤0.05);而边缘叶中P含量有升高,但与正常组相比无显著性差异。在下丘脑中,造模之后GABA及Glu含量与正常组相比显著降低(P﹤0.01,P﹤0.05),用药后,GABA含量显著升高(P﹤0.05),而Glu含量有上调趋势但无显著性差异;边缘叶中两种氨基酸含量均极显著降低(P﹤0.01),用药后,两者显著升高(P﹤0.05);额叶皮层中两种氨基酸造模之后均升高(P﹤0.01,P=0.0551),用药后,GABA含量显著降低(P﹤0.05),而Glu含量有降低趋势,但无显著性差异;造模组海马中两者含量显著升高(P﹤0.01),给药后,GABA含量显著降低(P﹤0.01),而Glu无显著变化。 结论:大鼠血清及四个脑区中P及GABA、Glu含量变化与PMS肝气逆证关系密切。中药白香丹胶囊对该病证模型大鼠血清与上述脑组织内P、GABA及Glu含量有不同程度的调节,这是该药治疗PMS肝气逆证的重要作用机制之一。血清及四个脑区中P与GABA、Glu含量变化可能是导致PMS肝气逆证的重要病因之一。
