5种稻田常用杀虫剂对斑马鱼的毒性及亚致死效应

摘要
采用半静态法在室内测定了5种杀虫剂氟虫腈、毒死蜱、三唑磷、杀虫单和仲丁威对斑马鱼的单一急性毒性,根据《化学农药环境安全评价试验准则》中农药对鱼类的毒性等级划分,评价了这5种杀虫剂对鱼类的安全性。结果表明:氟虫腈和毒死蜱96h-LC50在0.1~1.0mg·L-1之间,属于高毒级:三唑磷96h-LC50为1.0~10 mg·L-1之间,属于中毒级;杀虫单和仲丁威96h-LC50>10 mg·L-1,属于低毒级。 另外,在完成单剂急性毒性后,采用相加指数法相继进行三唑磷、杀虫单、仲丁威分别与氟虫腈复配及三唑磷、杀虫单、仲丁威与毒死蜱复配后对斑马鱼的联合毒性,结果表明仲丁威分别与氟虫腈和毒死蜱复配后对斑马鱼联合毒性为协同作用;杀虫单分别与氟虫腈和毒死蜱复配后,对斑马鱼的联合毒性先表现为拮抗作用,然后再表现为协同作用;三唑磷分别与氟虫腈和毒死蜱复配后,表现为拮抗作用。 研究3个亚致死浓度的单剂和复配剂于暴露96h期间对斑马鱼乙酰胆碱酯酶(AChE)、P450酶系7-乙氧基-3-异吩唑酮-脱乙基酶(EROD)和谷胱甘肽S-转移酶(GST)的影响,检测这3种生物标志物随不同农药暴露浓度和时间的变化趋势,探讨这些指标用于检测农药污染的可行性,从实验结果分析可以看出,斑马鱼乙酰胆碱酯酶AChE对毒死蜱、三唑磷和仲丁威暴露有良好的指示作用,EROD对这5种杀虫剂也有较好的指示作用。然而,斑马鱼GST的指示作用显得并不突出,各暴露浓度的氟虫腈对斑马鱼GST活性无明显影响;斑马鱼GST活性仅在最高暴露浓度的毒死蜱和三唑磷中才被显著诱导;GST活性随杀虫单浓度及时间变化较为复杂;高暴露浓度的仲丁威对斑马鱼GST却产生抑制作用。
