骨碎补抗骨质疏松活性部位的化学成分研究

摘要
槲蕨(Drynaria fortnei(Kunze)J.sm.)为水龙骨科植物,其根茎是中药骨碎补的主要来源之一,主要产于浙江、福建等地。中医辨证施治用于治疗各种原发性骨质疏松症,用以补肾填精,壮腰健骨。其主要成分是以柚皮苷为主的二氢黄酮类化合物,也足骨碎补活血祛淤、接骨疗伤和增强心肌细胞机能的主要活性成分,药用价值很高。 通过文献调研,我们发现以往对于本药材植物化学成分的研究主要集中在极性很小的脂溶性成分上,而其他部分的研究报道很少。本课题利用去卵巢小鼠动物实验和体外UMR106细胞增殖实验确定了骨碎补总提取物的活性部位。利用硅胶柱色谱,大孔吸附树脂柱色谱、Sephadex LH-20柱色谱、ODS开放柱色谱、制备Rp-HPLC等手段,从骨碎补60%乙醇提取物大孔树脂50%甲醇水洗脱部分中分离得到15个化合物。综合运用化合物的理化性质及IR、UV、GC-MS、ESI-MS、一维、二维核磁共振谱等光谱学方法确证其结构,分别为:5-乙氧基-2-羟基苯乙酯(1);北美圣草素(2);山柰酚3-O-α-L-吡喃鼠李糖苷(3);山柰酚-7-O-α-L-呋喃阿拉伯糖(4);3-乙酰胺基-4-羟基苯甲酸(5);原儿茶酸(6);江户樱花苷(7);紫云英苷(8):柚皮苷(9);5,7-二羟基色原酮-7-O-α-L-鼠李糖基-(1→2)-β-D-葡萄糖苷(10);山柰酚-3-O-α-L-鼠李糖基-7-O-β-D-葡萄糖苷(11);金鱼草素-6-新橙皮糖苷(12);5,7,3',4'-四羟基二氢黄酮-7-O-α-L-鼠李糖基-(1→2)-β-D-葡萄糖苷(13);4-O-β-D-吡喃葡萄糖基反式咖啡酸(14);β-谷甾醇(15)。 在分离得到的15个化学成份中,化合物1~8,10~14为首次从该属植物中分离得到的。共分离得到黄酮类化合物10个。
