盐酸氨溴素及地塞米松对脂多糖诱导大鼠急性肺损伤炎性细胞信号转导影响的实验研究

摘要
目的 通过腹膜腔注射脂多糖(lipopolysaccharide,LPS)诱导的大鼠急性肺损伤(acute lung injury,ALI)模型,并以盐酸氨溴素(ambroxol,AMB)和地塞米松(dexamethasone,DEX)两种药物作为干预因素,旨在探讨:(1)NF-κB信号转导与LPS诱导的急性肺损伤的关系;(2)AMB对NF-κB信号转导的调控作用;(3)DEX对NF-κB信号转导的调控作用;(4)AMB与ALI之间是否存在剂量依赖效应;(5)抑制NF-κB活性对LPS诱导炎症介质表达的作用;以及(6)联合应用AMB+DEX其作用是否相加等问题。 方法 1.实验动物分组:将207只健康S-D雄性鼠,分成54组,其中的27组,每组4-5只,分别设立对照组、AMB组、DEX组、LPS组、LPS+AMB组(AMB5、10、20mg/kg)、LPS+DEX组,以及LPS+AMB+DEX组,并根据时间的不同,即1、2、4hr,上述各组分别再设立3组;另设立病理组27组,每组3只,分组方法与前者相同。 2.动物制作模型:A)对照组:将生理盐水(N.S) 0.5ml注入腹膜腔,1hr后再射N.S 0.5ml;B)LPS组:先注射N.S 0.5ml入腹膜腔1hr后,再注射LPS(5mg/kg)入腹膜腔;C)AMB组:先注射AMB(10mg/kg)1hr后,再注射N.S 0.5ml;D)DEX组:先注射DEX(1mg/kg)1hr后,再注射N.S 0.5ml;
