本文以杂交松(Pinus elliottii Engelm ? Pinus caribaea Morelet)幼胚、成熟合子胚、实生苗的茎尖、子叶和茎段为外植体,进行器官发生和体细胞胚胎发生研究,探讨了不同因素对器官发生或体胚发生的影响。主要结果如下: 1 器官发生的研究结果表明:杂交松成熟合子胚与实生苗的茎尖是适合器官发生的外植体,最适合不定芽诱导的培养基是含盐量比较低的DCR与WPM。在WPM+BA0.25mg/L时,不定芽的诱导率达到最大值84%,在DCR+IBA0.5mg/L+BA0.1mg/L 的培养基中不定根的诱导率达到最大值56%。茎尖在1/2DCR+ BA0.5mg/L的培养基上,针叶基潜伏芽的诱导率可以达到85%,在BA 为1.0mg/L的DCR培养基上平均芽数达到最大值3.75。再生小苗的移栽成活率达到80%以上。NAA 不宜用于杂交松器官发生研究,而活性炭对器官发生则极为有利。同时发现针叶基潜伏芽具有一定的增殖能力。 2 体细胞胚胎发生的研究结果表明:单一细胞分裂素的培养效果优于多种细胞分裂素结合的培养效果,合子胚的成熟程度对试验结果的影响较大。不同杂交组合对试验的影响较大。B02? 河5 与B102? B2 的愈伤诱导率在DCR+BA1.0mg/L +2,4-D2.0mg/L 中的达到最大值95%,A05? L01 与B08? L09 在DCR+BA1.0mg/L+2,4-D5.0mg/L 的条件下,愈伤组织的诱导率最高,达到71.4%。因时间有限,没有系统地研究杂交松体胚发生的完整过程,如何促进愈伤组织的分化及体细胞胚的诱导有待进一步的深入研究。
