龟甲胶、鹿角胶对豚鼠骨关节炎软骨细胞内JNK、P38 MAPK表达的影响

摘要
目的:通过观察龟甲胶、鹿角胶含药培养液对豚鼠骨关节炎软骨细胞的增殖及JNK、P38 MAPK表达的影响,进一步明确龟甲胶、鹿角胶调控豚鼠关节软骨细胞退化的作用机制,为阐述中药通过干预关节软骨细胞JNK、P38 MAPK表达以防治骨关节炎提供新依据。方法:1.将实验分为4组(龟甲胶组、鹿角胶组、盐酸氨基葡萄糖组和空白组,分别简单记为G组、L组、A组和K组),制备各组含药培养液。2.对3月龄豚鼠OA软骨细胞分离、培养及鉴定,并建立软骨细胞体外培养体系。3.MTT比色法观察4组中各标记含药培养液分别对豚鼠F2代OA软骨细胞增殖的影响。4.Western-Blot检测各组豚鼠OA软骨细胞内JNK、P38 MAPK的表达量。5.QPCR测定各组豚鼠OA软骨细胞内JNK、P38 MAPK的mRNA含量。结果:1.Ⅱ型胶原免疫组化染色和甲苯胺蓝染色均为阳性,提示成功获取并建立豚鼠关节软骨细胞体外培养体系。2.MTT比色法结果:龟甲胶、鹿角胶和盐酸氨基葡萄糖含药培养液对豚鼠OA软骨细胞均有促增殖作用,效果为G组>L组>A组>K组,且P<0.05。3.Western-Blot法结果:龟甲胶、鹿角胶和盐酸氨基葡萄糖含药培养液均能有效抑制JNK、P38 MAPK的表达,其抑制效果为G组>L组>A组>K组,且P<0.05。4.QPCR结果:龟甲胶、鹿角胶和盐酸氨基葡萄糖含药培养液均能有效下调JNK、P38 MAPK的基因表达,且下调效果为G组>L组>A组>K组,P<0.05。结论:龟甲胶、鹿角胶通过下调豚鼠OA软骨细胞JNK、P38 MAPK mRNA的表达和JNK、P38 MAPK的表达,减轻豚鼠OA软骨细胞的退变,延缓豚鼠骨关节炎的病理进程。
