金银花中新绿原酸对LPS诱导的RAW264.7细胞的抗炎作用及机制

摘要
金银花作为一味传统中草药,最早记载见著于《神农本草经》。其初开或干燥花蕾可以作药,具有清凉解毒、祛风除热之功效。研究发现金银花中拥有大量的萜类、黄酮类、有机酸类等物质,具有散热抗炎、降低血脂、抗病毒、利胆、止血抗氧化等功效。作为咖啡酸类衍生物的绿原酸已被充分研究,但其同分异构体新绿原酸的研究却少见报道。本实验以LPS诱导的RAW264.7小鼠巨噬细胞体外炎症模型为载体,主要来探讨一定浓度范围和作用时间内新绿原酸的抗炎效果及作用机制。本论文的主要内容包括:首先采用不同浓度的药物作用于细胞,观察细胞生长状态;采用CCK-8试剂评估药物毒性,摸索出最适作用浓度;用Griess Reagent试剂检测新绿原酸对NO含量变化的作用;其次采用ELISA检测新绿原酸对PGE2生成的影响;同时采用实时荧光定量法(RT-qPCR)检测iNOS、COX-2、TNF-α、IL-1β IL-6 mRNA的表达;此外Western blot被用来检测COX-2和iNOS蛋白的表达以及NF-Kb和MAPKs炎症信号通路的变化。实验结果发现:在50-200μg·mL-1药物浓度作用时,药物对细胞无毒性;在200μg·mL-1时,与模型组相比,新绿原酸对LPS诱导RAW264.7产生的NO和PGE2的量均具有明显抑制作用(P<0.01),同时也显著下调iNOS和COX-2 mRNA和蛋白的表达(P<0.01);在100、200μg·mL-1浓度时,新绿原酸对TNF-α、IL-1β、IL-6 mRNA的表达量均有所下调(P<0.05);并且在200 μg·mL-1浓度时对NF-Kb通路中P-p65具有明显抑制作用。实验结果说明新绿原酸能抑制NO和PGE2的表达量,并对iNOS和COX-2蛋白和核酸均有明显抑制,对细胞因子TNF-α、IL-1β和IL-6核酸的表达也有显著下调作用。表明新绿原酸对由LPS诱导的RAW264.7细胞具有明显抗炎作用,并且其机制可能与下调P-NF-κbp65有关。
